I usually just have set ad spots and then find the program that works best in that spot instead of vice versa. Also on a related not, I <3 Rubicon (http://www.rubiconproject.com/website)

I have not. simply because I'm not focused on monetization.

I do experiment with ads, that's it. Their location is chosen with a purpose not to obstruct or disturb viewer experience on the site.

Quite often the money-making sites are also the ones that have ads at odd places which I find it irritating.

Yeah I have done a little ad placement testing, but not long enough to really figure anything solid out.

I have done some testing, and have so far found a pretty solid setup. Clearly, one of the best performing ads are those placed within the content itself, but that is annoying to regular readers. So, I use a snippet of code to detect a search referrer, and if true, it inserts an ad block into the content. This way, regular readers aren't annoyed, while a reader coming from search will see the ad and, those users are the most likely to click ads anyway, so it is a win-win.

It is no surprise that the inserted ad block for search referrers has more than double the CTR and eCPM of the standard ads :)

Yes, definitely have been playing around with ad placement, i agree with Jeremy, placing the ads within content is one of the best thing to do, i also find that placing an ad block (adsense) before my content works wonders but it's irratating to readers though.
